Chemotherapeutic treatment of neoplastic diseases is often restricted by adverse systemic toxicity, which limits the dose of drug that can be administered, or by the appearance of drug resistance. Lack of selectivity is only one (albeit a major) obstacle hindering the optimisation of drug effectiveness. Others include inaccessibility of target, premature drug metabolism and allergic reactions.
